烟草幼苗期耐低钾突变体的筛选及验证

摘 要:

为了探讨筛选烟草耐低钾突变体的最优条件, 以烟草野生型品种‘中烟100’为材料, 研究了不同钾离子浓度对其根生长的影响, 确定了在培养基中筛选耐低钾突变体的适宜钾离子浓度为0.075 mmol•L-1。然后以EMS诱变处理获得的‘中烟100’ M2代种子为材料, 进行了大规模的耐低钾突变体的筛选, 初步筛选出了99个M2代候选材料。选取50个材料继续进行了验证, 其中22个M3代突变体经复选鉴定弯根率≥75%, 这些突变体在低钾培养基上长势明显好于对照, 且绝大多数突变体的单位鲜重叶片的钾含量明显高于对照。研究结果可为烟草耐低钾突变体的筛选提供依据。

Screening and Identification of Low K+ Tolerant Mutants at the Seedling Stage of Nicotiana tabacum

Abstract:

In order to determine the optimal condition for screening of K+ tolerant mutants, a wild type variety, Nicotiana tabacum cultivar ‘Zhongyan 100’ was selected to analyze the effect of different concentrations of K+ on the growth of roots in this experiment and the optimal K+ concentration for screening of K++ tolerant mutants was considered to be 0.075 mmol•L-1. Then, M2-generation mutagenic seeds of ‘Zhongyan 100’ treated by EMS were selected for screening of K+ tolerant mutants. Finally, 99 M2-generation candidate individuals were obtained. We selected 50 individuals to further examine their tolerance to low K+. The results showed that 22 of 50 individuals were obtained with a higher root-bending rate (≥75%) and their growth vigor and K+ content of fresh weight per gramme was obviously better than that of the mutagenic parent, ‘Zhongyan 100’ in low K+ medium. This study can provide basis for screening of low K+ tolerant mutants in N. tabacum.
